Lentivirus 농축을 위하여 번거로운 초고속원심분리기를 사용하고 싶지 않을 경우 Lenti-X™ Concentrator를 사용하여 100 배까지 역가를 증가시킬 수 있으며 높은 MOI로 목적 세포에 감염시킬 수 있다.

간단한 조작과정 (Mix, Wait, Spin)
Lenti-X Concentrator는 빠르고 간단하며 효율적인 lentivirus 농축 방법이다. 실험과정은 lentivirus 상층액에 Lenti-X Concentrator 시약을 섞어 잠깐 동안의 반응 후 일반 원심분리기로 spin down 하는 3 단계로 매 우 간단하다(Figure 1). 초고속원심분리기를 사용하지 않고도 1 시간 이내에 100 배까지 역가를 증가시킬수 있다. Lenti-X concentrator는 Clontech의 Lenti-X Expression & Packaging System으로 만들어진 virus 뿐만 아니라 다른 모든 lentivirus 상층액을 농축할 수 있으며 원하는 부피와 농도로 쉽게 조절할 수 있다.
Lenti-X™ Concentrator 사용 방법
100 배까지 역가 상승
Lenti-X Concentrator 조작 과정을 사용하면 lentivirus를 본래 부피의 1/100 로 줄일 수 있고, 90%의 회수율로 107 ~ 109 IFU/ml로 역가를 상승시킬 수 있다 (Figure 2).
어떤 부피의 상층액으로도 유사한 결과를 얻을 수 있다.
초고속원심분리보다 훨씬 간단한 방법
Lenti-X Concentrator와 초고속원심분리를 이용한 바이러스 농축 방법을 비교하면 Lenti-X 법의 장점이 확연히 들어난다 (Table I). Lenti-X Concentrator 는 초고속원심분리법보다 더욱 유연하고 빠르며, 쉬울 뿐만 아니라 매우 효과적이다. 바이러스 상층액의 부피를 줄이거나 역가를 증가시켜야 할 때, Lenti-X Concentrator는 번거롭고 시간이 많이 소요되는 초고속원심분리 과정 없이 빠르고 간단하게 농축시킬 수 있다.

Lentiviral supernatant from a pLVX-ZsGreen1 vector was concentrated from 3 ml down to 30μl using the Lenti-X Concentrator Reagent and reflects a 100-fold increase in viral titer. The total amount of virus contained in each sample is also indicated, which shows that the resuspended pellet captured 90% of the virus present in original sample. Samples were titrated on HT1080 cells and analyzed by flow cytometry 48 hr post-transduction.
