LacZ & Beta-Gal Vectors and Assay Kit
Beta-galactosidase (the product of the LacZ gene)는 일반적으로 transfection 효율을 모니터하거나 promoter/enhancer 연구를 위한 리포터로써 이용되고 있다.
Clontech에서는 LacZ 유전자 활성을 검출할 수 있는 2가지 방법을 제공하고 있는데 ELISA reader를 이용하는 colorimetric assay와 luminometer를 이용하는
chemiluminescent assay 방법이 적용 가능하다.
- Colorimetric assay(비색분석법): Beta-Galactosidase Staining Kit은 포유류세포에서 beta-galactosidase의 발현을 colorimetric detection에
필요한 제품이 포함된 시스템이다. 이 Kit에는 beta-galactosidase (LacZ gene의 산물)가 X-gal의 가수분해를 촉진하여 파란색을 띄도록 한다. X-Gal Staining Mix를
이용하여 간편하게 세포의 고정, 세정과 인큐베이션이 가능하다. 그런 후 광학현미경으로 관찰하면 파란색으로 염색된 세포를 관찰할 수 있다.
- Chemiluminescent assay(화학적발광 분석법): Luminescent Beta-Galactosidase Reporter System 3 (vectors 포함; 이하 참고)과 Detection Kit
II (시약만 포함)는 화학적발광기질 chemiluminescent substrate 을 이용하여 beta-gal activity을 정량적으로 측정한다. 화학적발광 신호는 여러 시간동안
안정하며 scintillation counter나 luminometer 또는 x-ray film 감광을 통해 측정할 수 있다

- LacZ Vectors Clontech에서는 LacZ gene를 포함하는 3종류의 beta-gal mammalian reporter vectors를 제공한다.
유전자발현시 cis-regulatory elements의 영향을 분석하기 쉽도록 최대한 다양한 vector를 제공하고 있으며 이 vector는 transfection 효과를
normalize하는데 이용할 수 도 있다.
- pBeta-Gal-Basic (pLacZ-Basic으로 별도 판매된다.)는 진핵세포의 promoter와 enhancer sequences가 결핍되어 있다.
이 vector는 negative control로 이용하거나 promoter를 삽입하여 특성을 분석하는데 이용할 수 있다.
- pBeta-Gal-Control (pLacZ-Control로 별도 판매된다.)는 SV40 promoter와 enhancer sequences가 포함되어 있어 positive control로 이용하거나 promoters나 enhancer의 활성을 비교하는 reference로 활용할 수 있다.
- pCMV-LacZ는 human cytomegalovirus immediate early promoter (PCMV IE)로 beta-galactosidase를 고발현하는 mammalian reporter vector로 reference나 control plasmid로 이용할 수 있다. Beta-galactosidase 활성은 Luminescent Beta-Gal Detection Kit II으로 검출 할 수 있다.
- Complete Beta-Gal Reporter System Luminescent Beta-Galactosidase Reporter System 3는 pBeta-Gal-Basic와 pBeta-Gal-Control vectors, 그리고 chemiluminescent assay 시약을 포함하고 있다.
