pTet-tTS (Figure 1)는 tetracycline으로 조절되는 transcription silencer (tTS)를 발현시킨다. tTS는 KRAB silencing domain (SDKid-1)과 Tet Repressor protein (TetR)이 융합된 단백질이다 (1). tTS는 Tet-On Expression System에서 목적 유전자의 basal expression을 낮추어 준다. Doxycycline이 없을 때, tTS는 response plasmid의 TRE 부위에 있는 tetO 서열과 결합하여 유전자 발현을 방해한다. Clontech은 tTS를 발현하는 미리 제작된 다양한 종류의 cell lines를 제공하고 있다.
