제품설명
RetroNectin® (Recombinant Human Fibronectin CH-296)은 ① Cell-binding domain (C-domain, VLA-5와 결합) /  ② Heparin-binding domain (H-domain, 바이러스와 결합) / ③ CS-1 (VLA-4와 결합), 총 3종류의 기능성 단백질 단편을 가진 human fibronectin이다 (그림 1). 목적 세포가 VLA-4 또는 VLA-5를 발현하면 각각 CS-1 또는 Cell-binding domain과 결합하고 이때 H-domain에 결합한 바이러스와 목적 세포의 물리적 거리가 최소화되는 원리로 transduction이 촉진된다.

Fibronectin과 RetroNectin®의 구조 및 transduction 모식도 일반적으로 Hematopoietic cells 또는 그 외의 suspension 세포들은 transduction 효율이 낮지만 RetroNectin®은 낮은 독성으로 이런 세포에서도 고효율 transduction이 가능하다. 현재, RetroNectin®은 hematopoietic cells의 lentiviral / retroviral transduction에 gold standard로 사용되는 enhancer이며, 40건 이상의 임상 시험에 사용되어 임상 적용에도 큰 기대를 받고 있다.

목적세포로의 도입에는 (1) supernatant법 또는 (2) RBV (RetroNectin-bound virus)법을 적용할 수 있다.

(1) Supernatant 법
세포와 바이러스를 혼합하여 RetroNectin® 코팅 플레이트에 도입하는 방법으로 단시간에 간편하게 transduction을 진행할 수 있다 (목적 세포 첨가 후 plate를 원심분리하면 목적 세포와 바이러스의 상호작용을 촉진할 수 있다.).


(2) RBV (RetroNectin-bound virus) 법
바이러스 용액에 packaging cell-line에서 분비된 proteoglycan이나 envelope protein이 혼입되는 경우 transduction을 저해하는 경우가 있다. RBV법은 이러한 저해물질을 제거하고 transduction하는 방법이다. RetroNectin® 코팅 플레이트에 lentivirus 또는 retrovirus를 먼저 결합시키고 transduction inhibitor가 혼입된 배지를 제거한 후, 목적세포를 첨가하여 transduction한다. Supernatant법으로 충분한 transduction 효율이 나오지 않을 경우 RBV법을 추천하며 고농도로 바이러스를 사용하는 경우에서 높은 효율을 보인다 (바이러스의 titer가 부족한 경우 바이러스 용액 첨가 후 plate를 원심분리하면 바이러스의 transduction 효율을 더욱 높일 수 있다.).


* RetroNectin® Precoated Dish (Code T110A)는 RetroNectin®이 35 mm dish에 미리 코팅된 ready-to-use dish이다. 코팅과정을 생략하고 일정한 유전자 도입 효율을 얻을 수 있다.

* RetroNectin® GMP (Code T202)는 GMP 가이드라인에 준한 제조 및 품질 관리 공정에 의해 생산된 GMP grade의 RetroNectin®으로 연구용으로 판매되는 RetroNectin® (Code T100A/B)보다 고품질로 유전자치료나 세포 조제 등의 임상 연구에 사용 가능하다.

다양한 줄기세포에 세 개의 transduction enhancer를 이용하여 retrovirus를 사용한 형광 단백질의 발현 효율을 비교하였다. 그 결과, RetroNectin®은 GALV envelope receptor의 발현이 낮은 hCD34+, hADSC 같은 줄기세포에서도 높은 효율의 transduction이 가능하였다.

suspension cell인 SUP-T1에 lentivirus를 사용한 형광 단백질의 발현을 이용하여 transduction 방법 별 효율을 비교하였다. 그 결과 RetroNectin®을 사용한 Supernatant법 (static법, spin법), RBV-Spin법 모두 polybrene보다 높은 효율로 transduction이 가능하였다.

사용 상의 주의
- Freeze/thaw는 10회* 정도까지 가능하다.
* RetroNectin® GMP grade (Code T202)는 한번 융해 후 room temperature에서 12시간 이내 모두 사용되어야 한다.

비고
RetroNectin®은 lentivirus 또는 retrovirus의 유전자 도입 효율 증가 뿐만 아니라, T cell의 확대배양에도 적용 가능함이 확인되었다. 일반적으로 T cell 확대 배양은 Anti-CD3 antibody (OKT3)의 coating 후 배양액에 IL-2를 첨가하여 실시한다. 이때, RetroNectin®을 Anti-CD3 antibody와 함께 coating하면 기존의 확대 배양법 대비 약 1.5배 이상 확대 배양이 가능하고 높은 naive T cell 비율을 유지할 수 있다. 이러한 방법은 CAR-T cell 연구에 Gold standard로 사용되고 있다.
