EpiXplore Methylated DNA Enrichment Kit는 전체 genome의 hypo- & hyper-methylated DNA 모두를 enriching과 fractionating 하기 위한 간단한 magnetic bead 기반의 방법이다 (Figure 1). Enrichment는 sequencing 동안의 background를 감소시킴으로 유전자 조절 연구에 있어 최상의 결과를 가져다준다 (1).
EpiXplore Methylated Enrichment Kit를 선택해야 하는 이유
EpiXplore protocol은 methylated DNA와 특이성이 높은 Methylated DNA Binding (MDB) 단백질 사이의 affinity binding에 기반을 두고 있다.
  • 어떠한 antibody도 필요하지 않고 이후의 immunoprecipitation (IP) 절차가 요구되지 않는다.
  • 유연한 프로토콜은 (methylation 밀도차에 의한) fraction이나 한 번의 fraction으로도 methylated DNA를 elute하도록 해준다.
일반적인 IP-기반의 enrichment 방법 (MeDIP & MCIp)은 유연함이 부족하기 때문에 필요한 전처리과정을 행하면서 시작 물질의 감소가 발생한다. 좀 더 중요한 점은, MeDIP & MCIp은 methylated DNA의 gradient 분리가 이루어지지 않는다는 점이다.
왜 methylation 밀도에 의해 분리해야 하는가?
최근의 연구로 낮은 밀도의 methylation에 medium가 있는 genome 부분이 변화가 생기는 유전자 조절에서 중요한 역할을 한다는 것이 보고되고 있다 (2-4). EpiXplore Methylated DNA Enrichment Kit만이 enriched methylated DNA가 methylation 밀도에 기반을 둔 3개의 fraction으로 나누어지는 elution buffer를 포함하고 있다.
