Retro-X Q Vector는 고역가의 바이러스 생산과 신뢰성 높은 발현을 실현하고, promoter interference의 가능성을 최소화 하도록 설계되었다 (1).
본 Q Vector는 항생제 또는 형광 선별 마커 (Figure 1)와 함께 목적 유전자가 발현되고, 숙주의 genome으로 삽입된 후 5＇LTR의 promoter가 비활성되도록 설계되었다 (Figure 2). Self-inactivating vector는 promoter interference (2-5)의 가능성을 감소시켜 발현을 향상시킨다. MMLV LTR에서 transcript를 효과적으로 발현하지 못하는 세포에서도 확실한 발현을 보여준다(6).

Self-Inactivation Vector로 높은 바이러스 역가 생산.
5＇LTR의 CMV/MSV hybrid promoter는 packaging 과정 동안 높은 역가를 유도한다. 그런 다음 숙주 genome으로 삽입되는 동안 3＇LTR의 U3 영역의 결실이 5＇LTR에 복제되어 비활성화시킨다. 따라서 MCS의 바로 상류에 위치한 내부 CMV promoter에 의해서만 transcript가 발현된다. Vector set는 3개의 항생제 선별 마커 (Hyg, Pur, Neo)를 가지고 있으며, 이들은 목적 유전자와 함께 internal ribosome entry site (IRES)를 통하여 bicistron 형식으로 발현된다. LacZ control vector (pQCLIN) 또한 세트에 포함되어 있다.
