Clontech의 Retro-X Tet-On Advanced와 Retro-X Tet-Off Advanced Inducible Expression Systems은 Clontech의 최신 tetracycline 조절 유전자 발현 시스템과 효과적으로 유전자를 도입시키는 retroivirus가 결합된 제품이다 (1). 두 시스템 중 retroivirus vector는 Tet-Advanced inducible gene expression system에 다용도로 사용될 수 있으며, 광범위한 primary cell culture와 immortalized cell lines에도 사용될 수 있다. Cell lines를 만들어 놓으면, Dox의 농도 조절을 통해 시스템의 활성을 조절할 수 있다 (Figure 1).
Tet-Advanced System은 매우 높은 특이성으로 높은 발현을 나타내며, 여러가지 현상들에 영향을 받지 않는다.

Retroivirus Vector와 Packaging에 이상적

CalPhos Mammalian Transfection Kit (Cat. No. 631312)는 각 시스템에 포함되어 매우 효과적인 packaging cell transfection을 보증한다. 본 시스템은 Retro-X Universal Packaging System (Cat. No. 631530, 별매)과 함께 사용할 수 있기 때문에, 목적 세포에 숙주 지향성을 맞춘 바이러스를 매우 높은 역가로 생산한다. 또한, 목적 retrovirus를 cell line 이나 primary cell culture에도 효과적으로 도입 시킬 수 있도록 사용자가 직접 숙주 지향성을 선택할 수 있다. Vector는 packaging cell에서 고역가의 바이러스를 생산할 수 있도록 하는 Retro-X Q 기술을 특징으로 한다.
