Lenti-X Maxi Purification Kit는 정제를 거치지 않은 crude 상층액으로 부터 매우 많은 양의 바이러스를 정제할 수 있다. Filter를 이용한 정제 방법은 lentivirus 입자에 손상을 주거나 정제량을 감소시키는데 비하여 gravity column 방식의 실험조작으로 빠르고 간편하며 효과적이다. Gravity-flow column은 세척 과정 동안 column을 통해 부착되지 않은 물질들이 흘러가는 반면 상층액 내에 바이러스 입자들이 남아있게 되어, 정제된 바이러스는 온전한 형태와 기능을 회복한다. 본 시스템은 바이러스를 정제하는 과정이 plasmid DNA 정제만큼 간편하다.

왜 Lentivirus를 정제해야 하는가?

바이러스의 정제는 형질 도입하는 실험에 영향을 미칠 수 있는 세포 오염물질도 제거할 수 있다. 정제되지 않은 상층액은 잔존 plasmid DNA, 알려지지 않은 감염과 형질 도입 저해제, 세포 내 단백질과 혈청 단백질, 고도의 면역성 바이러스 단백질, 핵산, 바이러스 단편을 포함하고 있다. 민감한 목적 세포 또는 in vivo 적용을 위해서는 lentivirus 사용 전에 바이러스를 정제하여 이러한 오염물질을 제거하는 것이 필수적이다.
정제된 바이러스를 사용하면 ‘pseudo-transduction‘ 또한 방지할 수 있다.

간단한 정제 과정

Lenti-X Maxi Column은 사용하기 편리하도록 구성되어 있다. 시료 상층액 (9~45 ml)에 10x binding buffer를 첨가한 후 column에 넣으면 중력에 의해 시료와 buffer가 통과하고, 2 회 column 세척 과정 동안 불순물들은 제거되고 3 ml의 elution buffer로 정제된 바이러스를 회수한다.

Filter System보다 뛰어난 회수력과 순도

바이러스의 감염력을 유지하고 뛰어난 바이러스 회수능력을 발휘하는 것이 정제 과정의 핵심이다. 일반적으로 Lenti-X column은 시료에서 60% 이상의 바이러스가 정제되고 또한 감염력이 있다 (Figure 2). Anion exchange-based membrane system은 Lenti-X column 보다 낮은 회수능력을 보이며 종종 20% 이하인 경우도 있다. Lenti-X column은 filter system보다 더 높은 바이러스 순도와 매우 적은 단백질 검출을 보인다. Filter 정제 방식은 바이러스와함께 정제된 외부 단백질을 다량 포함하고 있다.
