제품설명

Lenti-X™ GoStix™ Plus는 Lentivirus 상등액으로부터 아주 빠르고, 간단하고 효율적으로 Lentivirus 역가를 측정할 수 있는 간이 역가 측정기이다. Lenti-X™ GoStix™ Plus를 이용한 역가 측정은 GoStix 카세트에 Lentivirus 상측액 20㎕를 가하고, 약 10분 후 Lentiviral p24의 여부를 밴드의 유무로 확인한다 (그림 1, 2). 이전 버전의 GoStix 제품에 비하여 Lenti-X™ GoStix™ Plus는 스마트폰 어플을 통하여 밴드 강도를 비교 및 수치화함으로써 렌티바이러스의 대략적인 역가를 간단하게 측정, 분석할 수 있다.
Lenti-X™ GoStix™ Plus 어플을 이용하여 도출된 역가 (GoStix Value 또는 GV)는 ELISA 방법의 결과와 유사하지만 ELISA 보다 더 쉽고 간단하게 역가를 측정(그림 6)할 수 있으며, 각각의 실험 (Batch)으로부터 나온 Lentivirus의 양을 비교 분석하고, 바이러스를 회수하는데 적절한 시간 (virus harvest time)을 결정하는 데에 최적이다 (그림 4).

Control band(C)는 항상 검출이 되며, Test band(T)가 나타났다는 것은 상층액에 충분한 Lentivirus가 존재한다는 것을 의미한다

Lenti-X Single Shots을 이용하여 ZsGreen1 Lentivirus를 2 반복 (duplicate)로 준비 후, 바이러스 회수시간에 따라 Infection unit과 GV Value (GV)를 측정하였다.

각기 다른 Lentiviral vector와 packaging system을 이용하여 ZsGreen1 Lentivirus를 제작 후, Infection unit과 GoStix Value(GV)를 측정하였다.

ZsGreen1 Lentivirus의 역가를 ELISA 방식의 Lenti-X p24 Rapid Titer Kit과 Lenti-X GoStix Plus로 측정하고 transduction 효율을 FACS로 분석하였다.
