제품설명

Lenti-X Integration Site Analysis Kit는 렌티바이러스 벡터가 host genome DNA에 provirus로 삽입된 부분을 GenomeWalker 기술을 기반으로 정확하게 분석하기 위한 제품이다. 렌티바이러스는 RNA genome virus로 DNA로 역전사 되어 host genome에 삽입된다. 바이러스 DNA는 host genome에 무작위로 삽입되기 때문에 목적유전자 및 endogenous 유전자 발현에 영향을 줄 수 있다. 따라서 provirus의 삽입부위의 식별은 host cell의 표현형 연구에 중요하다. 이 키트에는 실험에 필요한 모든 시약이 포함되어 있다. GenomeWalker 개요

1. 바이러스를 형질 도입한 세포에서 추출한 genomic DNA를 3종류의 blunt-end 제한효소 처리
2. 평활말단이 된 genomic DNA library에 GenomeWalker adaptor를 ligation.
3. Library에서 adaptor와 렌티바이러스 시퀸스가 포함된 부분을 PCR 증폭
4. Nested PCR을 통해 단일밴드 증폭, 증폭 된 산물은 sequencing을 통해서 integration부분 분석, 복수의 단편이 증폭되는 경우에는 각각의 band를 정제 한 후 sequencing을 통해서 분석
