제품 설명
Shasta™ Whole-Genome Amplification Kit은 Shasta™ Single Cell System에서 분리한 단일 세포로부터 고품질의 Whole-Genome Amplification(WGA) library를 제작할 수 있다. 이 시스템을 사용하면 완전 자동화된 프로토콜을 통해 한 번에 최대 1,500개 세포까지 단일 세포 DNA-seq library를 준비할 수 있다. 이 애플리케이션은 종양 이질성(tumor heterogeneity)을 이해하고, 비용을 절감할 수 있는 낮은 sequencing depth에서 copy number variation (CNV) 및 single nucleotide variation (SNV) 프로파일링을 통해 tumor subclonal analysis을 수행하는데 도움이 될 수 있다.

①Shasta™ Single Cell System으로 5,184 nanowell chip에 single cell 분주함. 이미징 소프트웨어를 이용하여 single cell이 포함된 well을 선택하고, ②Shasta™ Single Cell System은 preamplification 과 amplification steps에 필요한 시약을 분주하고, ③72개의 i5 및 72개의 i7 primer를 분주하여 on-chip barcoding PCR을 한다. ④off-chip purification후에 pooled & barcoded libraries는 Illumina® sequencing이 가능하다. 마지막으로 ⑤sequencing과 data analysis를 수행한다.

Library sequencing은 NextSeq 500/550 High Output Kit (150 cycles) (read length 2 x 75 bp)을 사용하여, PhiX spike in 18.81%로 실시하였다. Read 중 96.56%가 바코드 부착에 성공하였고, 바코드가 부착된 단일 세포의 경우, adaptor trimming 후 바코드가 부착된 read 중 99.12%가 unique read였으며, 바코드가 부착된 read의 94.06%가 hg38 reference genome 고유하게 매핑되었음을 확인하였다.
