제품설명

CRISPR/Cas9 genome editing (크리스퍼 게놈편집)은 타겟 유전자에 하나의 allele (monoallelic) 또는 두개의 allele (biallelic) 변이를 생성하는 것으로 알려져 있다. Guide-it Genotype Confirmation Kit (Code 632611)는 Genome Editing 후의 세포 클론에서 변이 유전자를 간편하게 Genotyping할 수 있는 제품으로, 한쪽 allele 변이 (monoallelic), 대립 allele 변이 (biallelic)를 구분할 수 있을 뿐만 아니라, Wild type (unchanged) 클론까지 구분할 수 있다.
Cas9과 single guide RNA (sgRNA)를 처리하여 게놈편집을 시행한 세포에서 타겟유전자를 direct PCR하여 증폭된 증폭산물 (amplicon)을 tube 내에서 재조합 Cas9과 sgRNA를 처리하여 in vitro cleavage를 수행한다. 그 후, 절단산물을 Agarose gel 전기영동을 시행함으로써 genotype을 판정할 수 있다.

* 본 제품에는 sgRNA를 합성하는 시약이 포함되어있지 않습니다. In vitro sgRNA 합성은 Guide-it™ sgRNA In Vitro Transcription Kit (Code 632635)을 이용하십시오.
