그림 1. 타사 대비 높은 도입효율과 낮은 독성을 보이는 TransIT^®-mRNA Transfection Kit. TransIT^®-mRNA Transfection Kit, Lipofectamine^® RNAiMAX (Life Technologies), Stemfect™ RNA Transfection Kit (Stemgent) 세가지 시약을 사용해 14일간 동일한 population의 BJ fibroblasts에 RNA를 매일 transfection하였다 (capping 및 polyadenylation이 완료된 EGFP mRNA - pseudouridine과 5mC modified bases (Trilink Biotechnologies, Inc.) 사용). 실험은 12-well plate에서 1 ㎍ RNA를 각각 시약의 최적 비율로 transfection하였고, transfection 후 매일 같은 위치에서 GFP와 위상차 이미지를 촬영하였다. Transfection 효율은 14회의 daily transfection 후 flow cytometry (Guava^® easyCyte™ 5HT)를 사용해 측정하였고 (blue bar), 세포 생존율을 flow cytometry를 통해 cell count로 측정하였다 (black line grey bar). error bar는 3 반복 실험한 well의 표준편차를 의미한다. 그 결과 TransIT^®-mRNA Transfection Kit는 14일간의 장기간의 반복되는 실험에서 높은 도입효율과 낮은 독성으로 타사 대비 높은 효율을 보였다.

Luciferase mRNA를 다양한 Cell에 도입 후 확인한 high level luciferase expression 결과. 12-well plate에 배양된 세포에 capping 및 polyadenylation이 완료된 luciferase encoding mRNA를 TransIT^®-mRNA Transfection Kit로 transfection 한 결과이다. Transfection 후 약 18시간 뒤 세포를 회수해 well당 total luciferase activity를 측정하였다. 그 결과 다양한 cell에서 높은 효율로 유전자를 발현시킬 수 있었다.

Transfection 후 약 18 시간 뒤 이 세포들을 Beta-gal Staining Kit로 염색해 LacZ가 transfection된 세포를 확인했다. 그 결과 LacZ mRNA를 도입한 cell만 효과적으로 스크리닝 할 수 있었다.
