TransIT®-2020 Transfection Reagent는 고효율, animal origin free의 transfection 시약으로 plasmid DNA를 다양한 mammalian cell에 도입할 수 있다. 이 제품은 primary cell 같은 일반적으로 transfection이 어려운 cell type 에서도 높은 level의 gene expression이 가능하며, transient transfection과 stable transfection 모두 적용이 가능하다. Animal-origin free에 serum compatible이 특징으로 transfection 후 별도의 media change가 필요하지 않다는 장점이 있다.

그림 1. 타사제품 대비 높은 도입효율과 낮은 세포독성을 나타내는 TransIT®-2020 Transfection Reagent. Human umbilical vein endothelial cells (HUVEC)에 각각의 시약과 DNA를 위 막대 그래프에 명시된 비율 (시약 : DNA)로 처리하여 luciferase expression plasmid를 도입하였다. 96-well plate에서 각 well에 0.1 ㎍의 plasmid DNA를 transfection 하였고, 24시간 뒤 luciferase activity (막대 그래프)와 lactatedehydrogenase (LDH) level (선 그래프)을 측정하였다. LDH level은 시중에 판매되는 colorimetric assay 제품으로 분석되어 transfection 하지 않은 세포 대비 % cytotoxicity로 정리되었다. 그 결과 TransIT®-2020 Transfection Reagent는 높은 도입효율과 낮은 독성으로 타사 대비 높은 효율을 보였다.


그림 2. 다양한 cell type 에서의 뛰어난 유전자 발현. 96-well plate에서 각각의 cell type에 luciferase expression plasmid (0.1 ㎍/well)를 산업에서 대중적으로 사용되는 프로토콜로 transfection한 결과이다. 이때 reagent와 DNA의 비율은 cell-type 별로 최적화되었다: TransIT®-2020 (Mirus Bio, 2:1 or 3:1), FuGENE® HD (Roche, 3.5:1), Lipofectamine® 2000 (Life Technologies, 1.5:1, 3:1 or 5:1). Transfection 후 24시간 뒤 luciferase 활성을 측정하였고, TransIT®-2020을 사용한 경우의 luciferase 활성을 기준으로 normalize하였다. 그 결과 TransIT®-2020 Transfection Reagent는 다양한 cell type에서도 고효율로 유전자를 발현시킬 수 있었다.
