그림 1. TransIT®-X2 System은 다양한 cell type에서 고효율로 유전자 발현 가능. TransIT-X2® Dynamic Delivery System과 Lipofectamine® 2000 Transfection Reagent를 사용해서 luciferase를 encoding하는 plasmid DNA를 41개의 다른 cell type에 3 : 1 (reagent : DNA)의 비율로 transfection 하였다. Transfection 24시간 후에 standard luciferase assay를 진행한 결과 TransIT-X2®를 사용하는 경우 41개 중 36개의 cell type에서 luciferase 활성이 동등하거나 더 높았다. 이 중 17개의 cell type에서 TransIT-X2® 가 2배 이상 높았고 해당 cell type을 ‡로 표시하였다.
제품 설명
TransIT-X2® Dynamic Delivery System은 non-liposomal polymeric 시스템으로 primary cell을 포함한 다양한 cell type에서 고효율 transfection이 가능하다. 이 제품을 사용하면 DNA, siRNA/RNAi 및 DNA와 siRNA 등의 co-transfection을 진행할 수 있다. 또한 serum compatible transfection이 특징으로 transfection 후 별도의 media change가 필요하지 않다는 장점이 있다.

그림2. 타사제품 대비 높은 transfection 효율과 낮은 세포독성을 나타내는 TransIT-X2® Dynamic Delivery System. A549 (A)와 MDCK (B) 세포에 luciferase를 encoding하는 plasmid를 TransIT-X2®, Lipofectamine® 2000 (Thermo Fisher Scientific), Lipofectamine® 3000 (Thermo Fisher Scientific)으로 각각의 최적 조건에서 24시간 동안 transfection 한 결과이다. Luciferase activity로 transfection 효율을 측정하고, 손상된 세포의 세포질에서 방출되는 LDH를 정량해 세포독성을 평가하였다. 그 결과 Lipofectamine® 2000 및 Lipofectamine® 3000에 비해 TransIT-X2®에서 높은 transgene 발현율과 낮은 세포독성을 확인할 수 있었다.


그림 3. 다양한 cell-line 및 primary cell에서 고효율 GFP 발현. A549, CHO-K1, Hep G2, MDCK, LNCaP, PC-12, primary human mammary epithelial cells (HMEC), normal human dermal fibroblasts (NHDF) 세포에 EGFP를 encoding하는 plasmid를 TransIT-X2® Dynamic Delivery System로 transfection 한 결과이다. 96-well plate에서 0.2~0.4 ㎕의 TransIT-X2®로 0.1 ㎍의 DNA를 transfection 했고 (2:1, 3:1, 4:1 / reagent: DNA ratio), 48시간 뒤 3반복으로 guava easyCyte™ 5HT Flow Cytometer에서 분석되었다. 그 결과 TransIT-X2® Dynamic Delivery System은 다양한 cell-line 및 primary cell에서 고효율로 GFP를 발현시킬 수 있었다.

그림 4. 타사제품 대비 RNP의 transfection 효율이 높은 TransIT-X2® Dynamic Delivery System. HEK293T/17과 U2OS 세포에 2-part gRNA (IDT)를 타겟하는 PPIB (cyclophilin B)와 Cas9 protein (PNA Bio)으로 제작한 Ribonucleoprotein (RNP) complexes를 TransIT-X2® Dynamic Delivery System (1 ㎕/well) 또는 Lipofectamine® CRISPRMAX™ (1.5 ㎕/well and 1 ㎕/well of Lipofectamine® Cas9 Plus™ Reagent, ThermoFisher), Lipofectamine® RNAiMAX (1.5 ㎕/well), Lipofectamine® 3000 (1.5 μl/well and 1 μl/well of P3000™ Reagent, ThermoFisher)로 제조사의 프로토콜에 따라 transfection 한 결과이다. 6 nM Cas9 protein (PNA Bio)과 다양한 양의 gRNA (6 nM, 12 nM)로 테스트되었고, transfection 48시간 후 T7E1 mismatch detection assay로 cleavage efficiency를 측정하였다. 그 결과 TransIT-X2® Dynamic Delivery System은 타사 대비 높은 효율로 RNP를 도입하여 고효율로 유전자 편집을 진행할 수 있었다.
