제품설명
DEF-CS™ 500 Culture System은 기존의 Colony 배양 방식이 아닌, feeder free환경에서 monolayer 형태로 간편하게 iPSC/ESC 확대 배양이 가능한 배양 시스템이다.
DEF-CS™ 500 Culture System에서 배양된 iPSC/ESC는 자연분화 없이 강력한 미분화를 유지하기 때문에, 배양 과정 중 분화된 세포를 제거하는 과정이 불필요하다. 또한, 본 system배양 하에서는 iPSC/ESC의 성장률을 효율적으로 예측 가능하기 때문에, 뛰어난 재현성으로 균일한 확대배양을 할 수 있다. DEF-CS™ 500 Culture System에서 배양된 iPSC/ESC는 single cell cloning 효율이 뛰어나기 때문에, iPSC/ESC의 다양한 single cell application (예: 유전자 편집)에 최적이다.

각 배지에서human iPS 세포주 (253G1)를 배양한 (계대 (passage) 후 5일 배양) 사진이다. 배양 조건은 각 제품의 권장 프로토콜에 따라 진행하였다. DEF-CS™ 500 Culture System는 다른 배지와 달리 colony를 형성하지 않고, monolayer 형태로 배양되는 것을 확인할 수 있다.

human iPS 세포주 (253G1)를 각 배지의 권장 프로토콜에 따라 5 주간 배양 후 회수한 세포에서, 미분화 마커(TRA1-60, SSEA4)의 발현을 flow cytometry로 분석했다. DEF-CS™ 500 Culture System에서 배양된 iPSC는 강력한 미분화를 유지하는 것이 확인되었다.

DEF-CS™ 500 Culture System에서 hiPSC (ChiPSC18)를 20회 계대배양 후 karyotype (핵형)을 분석한 결과 정상적인 핵형이 유지되고 있음을 확인하였다.

DEF-CS™ 500 Culture System를 사용하여 1 well 당 한 개의 GFP-actin hiPSC를 배양하였다. 배양 24시간 후 (그림 A,B)와 배양 16일 후 (그림 C, D, E, F) 세포 형광 사진을 확인해본 결과, 하나의 세포에서 다수의 세포로 균일하게 확대배양 된 것을 확인할 수 있었다.
