제품설명

Cellartis Definitive Endoderm Differentiation Kit는 인간 iPS 세포로부터 내배엽 (Definitive Endoderm : DE) 세포로 플레이트 배양 상태에서 분화를 유도하기 위한 완전한 배지와 ready-to-use 코팅용액으로 이루어진 제품이다. 이 제품에는 분화 유도 전에 미분화 상태에서 iPS 세포를 배양 할 수 있는 배지 Cellartis DEF-CS 100 Culture System도 포함되어 있다. 본 제품에 의해 얻어진 내배엽(DE) 세포는 간세포와 췌장 내배엽 세포로의 분화 유도에 사용할 수 있다 (참고 문헌 1). 내배엽(DE) 세포에서 간세포로의 분화는 Cellartis Hepatocyte Differentiation Kit (Code Y30050)의 사용을 권장한다. 또한 Cellartis iPS cell to Hepatocyte Differentiation system (Code Y30055)을 사용하면 iPS 세포로부터 간세포까지의 분화가 가능하다.

2종의 인간 iPS 세포주 (ChiPSC18, P11025)를 본 제품을 이용하여 내배엽 세포로 분화 유도하고 내배엽 마커 유전자의 mRNA 발현량을 RT-qPCR을 이용하여 분석했다. 미분화 세포(day0)에서 내배엽 세포로 분화된(day7) 것을 확인할 수 있었다.
