제품설명

TransIT-VirusGEN^® Transfection Reagent는 expression vector와 packaging plasmid를 HEK293 타입의 패키징 세포로 효율적으로 도입하여, AAV 또는 Lentivirus의 생산율을 높여주는 transfection 시약이다.
그 중에서도 본 VirusGEN^® LV Transfection Kit 제품은 TransIT-VirusGEN^® Transfection Reagent를 기반으로 plasmid 도입 효율을 높이기 위한 VirusGEN^® LV Complex Formation Solution과 VirusGEN^® LV Enhancer가 추가되어 Lentivirus의 효율적인 생산을 위해 최적화된 제품이다.
VirusGEN^® SELECT LV Transfection Kit는 전임상 연구 및 공정 개발에서 필요한 대규모 바이러스 생산을 위한 최적의 시약으로 간편하고 비용 효율적인 실험 프로세스를 제공한다. 또한 전임상용 바이러스 벡터 제작을 위한 transfection 시약으로 사용할 수 있도록 품질 보증 시험을 완료하였다.

따라서, 임상 시험 또는 상업적인 바이러스 생산을 위해서는 VirusGEN^® GMP LV Transfection Kit를 사용할 수 있다.

Suspension 형태로 배양한 Freestyle 293-F세포 (FreeStyle F17 Medium에서 배양)(A)와 Adherent 형태로 배양한 293T/17세포(B)에 Trans IT-VirusGEN^® Transfection Reagent (reagent : DNA (vol : wt) = 3 : 1), 타사 B4 (2:1)(A), 3:1 (B), 타사 B5 (2:2:1 (A) 또는 3:3:1 (B)), 25 kDa PEI (4:1, Polysciences) 으로 transfection 하였다.
이때 pLKO.1-puro-CMV-TurboGFP™ transfer vector 와 Lentivirus Packaging Mix powered by MISSION^® (1:1 ratio, 1 µg/ml)을 이용하였다. Transfection 후 세포 배양 상등액을 회수하여 0.45 µm filtering 후, 293T/17 cell에 8 µg/ml TransduceIT™로 transduction하였다. 감염 72시간 후에 GFP 발현을 유세포측정기로 측정하여 functional titer를 확인하였다.
*Functional titer는 GFP 발현세포가 20% 미만인 virus 희석액으로부터 측정하였고, error bar는 3반복 실험군의 표준편차이다.

TransIT-ViruGEN^® Transfection Reagent, VirusGEN^® LV Transfection Kit, A제품 3, C제품 1을 이용하여 lentivirus의 생산 능력을 비교하였다. Expi293 Expression Media (Thermo Fisher Scientific)에서 배양한 Expi293F cells (Thermo Fisher Scientific)에 plasmid set (A. pALD-Lenti EGFP-A transfer vector, pALD-VSV-G-A, pALD-Rev-A와 pALD-GagPol-A packaging vectors (DNA비율 3:0.5:0.5:2, Aldevron), B. pLKO.1-puro-CMV-TurboGFP transfer vector (Millipore Sigma), ViraSafe Pantropic Packaging mix (pRSV-Rev, pCMV-VSV-G, pCgpV, CellBio Lab) DNA비율 3:0.5:0.5:2)을 유전자 도입하고 바이러스를 회수하였다. 회수한 바이러스를 293T/17세포에 감염시키고 72시간 후에 GFP의 발현을 확인하여 역가를 측정하였다.
그 결과, TransIT-VirusGEN^® Reagent, VirusGEN^® LV Transfection Kit를 이용했을 때 더 많은 역가의 lentivirus를 생산할 수 있었고, 특히 VirusGEN^® LV Transfection Kit를 이용하였을 때 가장 많은 양의 lentivirus를 생산할 수 있었다.
