X-¥á-Gal is a chromogenic substrate for yeast galactosidase (MEL1) and is used for detecting GAL4- based two-hybrid interactions directly on agar (Figure 1). This quick and easy assay eliminates the need for time-consuming ¥â-galactosidase liquid and filter-lift assays in two-hybrid analysis.

The X-¥á-Gal assay detects the activation of the yeast MEL1 gene, a GAL4-regulated gene used in two-hybrid analysis. MEL1 encodes the secreted enzyme ¥á-galactosidase, which hydrolyzes colorless X-¥á-Gal into a blue end-product. Yeast colonies that express ¥á-galactosidase in response to a positive two-hybrid interaction turn blue when grown on media containing X-¥á-Gal (Table 1).
