제품설명
TransIT-VirusGEN® Transfection Reagent는 expression vector와 packaging plasmid를 HEK293 타입의 패키징 세포로 효율적으로 도입하여, AAV 또는 Lentivirus의 생산율을 높여주는 transfection 시약이다.
VirusGEN® AAV Transfection Kit및 VirusGEN® LV Transfection Kit는 TransIT-VirusGEN® Transfection Reagent를 기반으로 plasmid 도입 효율를 높이기 위한 enhancer / complex formation 시약이 추가된 kit로 각각 AAV와 Lentivirus의 효율적인 생산을 위해 최적화된 제품이다.
TransIT-VirusGEN® SELECT Transfection Reagent는 연구용 시약인 TransIT-VirusGEN® 과 조성은 같지만 전임상용 바이러스 벡터 제작을 위한 transfection 시약으로 사용할 수 있도록 품질 보증 시험을 완료하였다. ([Example Certificate of Origin (PDF) | Example Certificate of Analysis (PDF) | Example BSE TSE Statement (PDF)])
TransIT-VirusGEN® SELECT은 PEI, PEIpro® reagent보다 뛰어난 성능으로, 간단하고 합리적인 비용으로 바이러스를 대량 생산할 수 있다.
TransIT-VirusGEN® GMP Transfection Reagent는 USP <1043>에 따라 세포, 유전자 및 조직 공학 제품 개발 시 보조시약으로 사용할 수 있어, 임상 시험 또는 상업적인 바이러스 생산 시 사용을 권장한다.
TransduceIT™ Transduction Reagent는 hexadimethrine bromide 로, 이를 사용하면 lentivirus 또는 retrovirus transduction 효율을 높일 수 있다.

Suspension 형태로 배양한 Freestyle 293-F세포 (FreeStyle F17 Medium에서 배양)(A)와 Adherent 형태로 배양한 293T/17세포(B)에 Trans IT-VirusGEN® Transfection Reagent (reagent : DNA (vol : wt) = 2 : 1), 타사 B4, 타사 B5, 타사 C3 (25kDa PEI)를 사용하여 pAAV-hrGFP, pAAV-RC, pAAV-Helper (1 : 1 : 1 ratio, 1.5 ㎍/㎖, Agilent Technologies)를 transfection 하여 바이러스를 생산하였다. 생산된 바이러스는 HT1080세포에 감염, 48시간 뒤 GFP발현을 유세포측정기로 측정하였다.
*Functional titer는 GFP 발현세포가 20% 미만인 virus 희석액으로부터 측정하였고, error bar는 3반복 실험군의 표준편차이다.

Suspension 형태로 배양한 Freestyle 293-F세포 (FreeStyle F17 Medium에서 배양)(A), Adherent 형태로 배양한 293T/17세포(B)에 TransIT-VirusGEN® Transfection Reagent (reagent : DNA (vol : wt) = 3 : 1), 타사 B4, 타사 B5, 타사 C3 (25kDa PEI)를 사용하여 pLKO.1-puro-CMV-TurboGFP transfer vector, MISSION® Lentivirus Packaging Mix (1 : 1 ratio, 1 ㎍/㎖)을 transfection 하였다. Transfection된 세포의 배양 상층액을 0.45 ㎛ 필터로 여과하여 8 ㎍/㎖TransduceIT™ 과 함께 293T/18에 감염시켰다. 72시간 뒤 GFP발현을 유세포측정기로 측정하였다.
