Illumina NGS library quantification by qPCR

제품설명

High-quality의 Illumina next-generation sequencing 데이터를 얻기 위하여, 적절한 양의 library DNA를 이용하는 것이 필수적이다. Library DNA의 양이 부족하면 cluster 밀도 (cluster density)가 낮아져 시퀀싱 수율(sequencing yield)이 낮아진다. 반면, Library DNA의 양이 너무 많으면 Cluster 밀도가 높아져 High-quality의 데이터를 얻을 수 없다. 전기영동(Electrophoresis)이나 분광 광도 측정(spectrophotometry)과 같은 기존의 NGS Library 정량 방법은 sensitivity가 낮고, adaptor-bound DNA에 대하여 비특이적 일뿐만 아니라, 많은 양의 Library DNA가 필요하다.
Library Quantification Kit은 고감도의 qPCR 기반의 정량법을 사용하기 때문에 Illumina의 NGS 반응에 사용되는 adaptor sequence를 특이적으로 검출할 수 있다. 또한, 샘플 내에 adaptor가 부가되지 않은 DNA분자가 있더라도 sequencing library의 농도를 측정 가능하며, library 제작 과정에서 adaptor ligation을 확인하는 데 사용할 수도 있다.

본 제품에는 standard curve 작성을 위한 DNA Standard가 포함되어있어, library DNA의 절대정량이 가능하며, DNA standards for Library Quantification(Code 638325)는 별도로 구매 가능하다.

Quantification of three lots of pre-diluted DNA Standards (0.01, 0.1, 1, and 10 pM) was performed by real-time PCR using Terra qPCR Direct SYBR Premix following the protocol in the Library Quantification Kit user manual. All reactions were performed in triplicate, and the resulting Ct (SDM) values for the triplicates were averaged. The variation between lots was <5%.
