ChIP Elute Kit은 chromatin immunoprecipitation (ChIP) 방법으로 얻어진 DNA를 회수, 정제할 수 있는 제품이다. 본 제품을 사용하면 Single-stranded DNA (ssDNA)를 약 1시간만에 회수할 수 있으며, 회수된 DNA는 DNA SMART ChIP-Seq Kit (code 634865)에 바로 적용할 수 있다.

DNA SMART ChIP-Seq Kit은 별도의 ligation 과정 없이(Ligation-independent manner), 목적 DNA 양 말단에 Illumina-specific sequencing adaptor를 부가하여 Illumina platform에 적합한 DNA Library를 제작하는 제품이다. Chromatin Immunoprecipitation(ChIP)으로부터 회수한 DNA는 양이 매우 적기 때문에 Chromatin Immunoprecipitation-sequencing(ChIP-Seq)에 적용하기가 매우 어렵다. DNA SMART ChIP-Seq Kit은 ligation 과정 없이, 100 pg - 10 ng의 소량의 DNA 샘플로부터 Illumina platform용 sequencing library를 제작할 수 있다. 또한 절단 또는 분해된 double strand DNA(dsDNA) 뿐만 아니라 single-strand DNA(ssDNA)도 이용할 수 있기 때문에 ssDNA로부터 dsDNA로 만들기 위한 별도의 과정이 필요 없다. 본 제품을 이용하면 약 4시간만에 효율적으로 library 제작이 가능하다.

본 제품은 Clontech 특허의 SMART 기술을 적용한 새로운 제품으로 DNA 주형으로부터 DNA를 합성할 수 있다 (DNA dependent DNA polymerase). DNA SMART ChIP-Seq Kit의 핵심은 극소량의 샘플 (low-input sample)로부터 ligation 과정 없이 Illumina용 sequencing adaptor를 부가할 수 있다는 점이다.

표 1. The DNA SMART ChIP-Seq Kit uses a combined size selection and clean-up step after library amplification by PCR. Compared to other protocols that perform size selection before PCR amplification, post-PCR size selection results in a higher yield while maintaining the quality of the libraries.

표 2. The DNA SMART ChIP-Seq Kit has the sensitivity to generate sequencing libraries from very small amounts of fragmented DNA. The number of unique, non-duplicate reads is high across all input levels, and the number of peaks identified is similar across input amounts.
