목적세포의 nuclear DNA에 삽입된 lentivirus의 양을 정량함으로써, lentiviral supernatant stock 내에 존재하는 실제 역가 (real titer, effective titer)와 MOI (Multiplicity of infection)를 보다 정확하게 측정할 수 있다. 이를 통하여 얼마나 많은 바이러스 유전체 (viral genome)가 목적 세포에 삽입이 되었는지를 예측할 수 있다.

- Lenti-X™ Provirus Quantitation Kit (Code 631239)는 TB green 기반의 qPCR 방법으로 transduced cell 내의 삽입된 렌티바이러스 (integrated lentivirus (provirus))의 copy number를 빠르게 정량할 수 있는 제품이다. 본 제품은 genomic DNA purification kit와 200회분의 TB Green® qPCR premix 및 calibrated control template를 포함하고 있다.

* 본 제품 내에 포함된 primer는 Packaging Signal에 인접한 HIV-1 genome 영역을 증폭할 수 있도록 디자인되어 있습니다. 본 제품은 다카라바이오에서 판매하는 모든 lentivirus vector 뿐만 아니라 일반적으로 사용되는 HIV-1 based lentiviral vector에 호환 가능합니다. 만약, 사용중인 vector와의 호환성을 확인하고자 한다면, 고객지원메일 (support@takara.co.kr)로 벡터서열(.txt)을 첨부하여 문의 바랍니다.

HT1080 세포는 AcGFP1 lentiviral expression vector를 사용하여 다양한 MOI로 감염시켰다. 감염 72시간 후 puromycin 항생제를 사용하여 selection 하였으며 1주일 후 안정적으로 transduction된 clone을 선별하여 증식 및 분석을 진행했다. AcGFP1 발현 수준은 flow cytometry를 통해 분석되었고 genomic DNA 내 provirus 함량은 Lenti-X™ Provirus Quantitation Kit (Code 631239)로 측정했다. 낮은 copy number (>3; n = 8) 또는 높은 copy number (>9; n = 7)의 두 그룹의 결과를 통합한 그림이며, provirus 함량이 높을수록 발현 수준이 높은 것을 확인할 수 있다.

Lentivirus stock (100 ㎕)에서 동일한 양을 사용하여 HT1080, HeLa, HEK 293 세포를 감염시켰다. 72시간 후 puromycin 항생제 selection을 시행했으며, 1주일 후, 안정적으로 형질도입된 세포 집단의 provirus copy 수를 Lenti-X™ Provirus Quantitation Kit (Code 631239)를 사용하여 측정하였다. 위 그래프는 동일한 양의 감염일지라도 세포 유형에 따라 다른 양의 provirus로 측정될 수 있음을 시사한다.
