Lenti-X™ T-Cell Transduction Sponge는 다공성 alginate (알긴산) 소재 스펀지로 microfluidics 기반 장비를 사용하지 않고도 T 세포에서 microfluidics 기반 lentivirus transduction 효율을 향상시킬 수 있는 혁신적인 기술이다. 시간이 많이 소요되는 spinoculation 방법이나 세포 viability 유지를 위해 최적화가 필요하고 세포독성이 문제인 화학적 형질도입 enhancer (polybrene 등) 없이 T 세포 activation 시약 (rhIL-2, anti-human CD3 & CD28 항체)이 최적화된 혼합비율로 스며진 스펀지 하나만으로, 2 x 10⁶~10⁷ 개의 세포를 1 step으로 T cell activation 과 transduction을 동시에 진행할 수 있다.

타겟 세포 (T cell 또는 PBMC)와 lentivirus를 혼합하여 스펀지에 분주하고 37℃에서 1 시간 동안 배양하여 혼합물이 스펀지에 잘 흡수되도록 한다. 흡수 후, 세포 배양 배지를 스펀지에 분주하고 37℃에서 48시간 동안 배양한다. 배양 후, 동봉된 Release Buffer를 이용해 스펀지를 녹이면 세포 viability에는 영향을 미치지 않으면서 transduction된 T cell을 쉽게 얻을 수 있고 이렇게 얻어낸 세포들은 추가로 확대배양 및 후속 실험에 사용할 수 있다.
본 제품의 사용자 친화적 프로토콜은 세포 handling 과정을 줄이고 적은 reaction 부피로 총 반응량을 줄여, 소량의 바이러스로도 실험이 가능하며 기존의 transduction 방식과 동등하거나 우수한 효율을 보인다.

4 x 10⁶ human primary T cell과 ZsGreen1을 발현하는 lentivirus를 다양한 MOI (multiplicities of infection)로 혼합하여 3개의 그룹으로 비교하였다. Lenti-X™ T-cell Transduction Sponge ("T-cell sponge")에 첨가한 그룹, CD3/CD28 ImmunoCult 시약 ("Conjugate")에 처리한 그룹, 처리하지 않은 그룹은 대조군 ("Unstim")이다. 48 시간 후 각 그룹의 세포에서 CD69 발현을 FACS 분석하였고, Conjugate 그룹보다 T-cell sponge 처리군에서 T cell activation 정도가 동등하거나 우수하였다.

4 x 10⁶ human primary T cell을 ZsGreen1을 발현하는 lentivirus를 일정한 MOI로 혼합하여 Lenti-X™ T-Cell Transduction Sponge ("T-cell sponge")에 첨가하였다. 다른 한쪽은 CD3/CD28 ImmunoCult 시약에 이틀 동안 activation한 후 polybrene (8 ㎍/㎖)과 같이 lentivirus를 넣어 1,500 g 90분 원심분리하였다 ("Spinoculation"). Transduction 효율은 그래프에 표시된 시점에서의 ZsGreen1 발현을 FACS 분석으로 평가하였으며, 표시된 백분율은 7일째의 GFP+ 발현을 나타낸다. 그 결과, Spinoculation보다 T-cell sponge 그룹에서 더 효과적으로 transduction되었음을 확인하였다.

4 x 10⁶ human primary T cell을 ZsGreen1을 발현하는 lentivirus를 일정한 MOI로 혼합하여 Lenti-X™ T-Cell Transduction Sponge ("T-cell sponge")에 첨가하였다. 다른 한쪽은 CD3/CD28 ImmunoCult 시약에 이틀동안 activation한 후 polybrene (8 ㎍/㎖)과 같이 lentivirus를 넣어 1,500 g 90분 원심분리하였다 ("Spinoculation"). 세포생존율은 transduction 후 그래프에 표시된 시점에서 7-AAD 염색 및 FACS 분석으로 평가하였으며, T cell sponge 를 이용한 그룹에서 더 높은 cell viability를 보였다.
