제품설명
Capturem™ His-tagged Purifcation 시리즈는 차세대 멤브레인 기술을 적용하여 His-tagged protein을 정제할 수 있는 혁신적인 제품으로써, High-capacity Ni-NTA Capturem™ membranes이
차용되어있다. 기존의 metal affinity resin에 비하여 보다 다양한 샘플 (박테리아, 포유류 적용 가능)과 다양한 조건 (비변성 또는 변성 조건 (8M urea, 6M Guanidine-HCl)에서도
적용이 가능하다.
Capturem™ 시리즈는 레진을 이용한 정제와는 달리 빠른 시간 내에 실온에서 조작하기 때문에 단백질의 분해(degradation), 응집 (aggregation), 실활 (loss of activity) 등의
단백질 열변성, 불활성화에 대한 위험을 감소시킬 수 있다. 또한 Bed volume이 작고 (2ul: Miniprep), Binding capacity가 높아 (
75 mg/cm3), 고순도, 고농도의 단백질을 정제할 수 있다.
Capturem™ His-tagged Purifcation 시리즈는 아래와 같이 Miniprep, Maxiprep, 24well plate, 96well plate, Large volume으로 제공된다 (표 1).

* Maxiprep Kit에는 50㎖ tube용, 24well plate, 96well plate에는 각각 24well plate, 96well plate용 centrifuge가 필요합니다.
** Large volume의 경우, Vacuum source (e.g. vacuum pump equipped with a trap and a vacuum controller, or a peristaltic pump)가 필요하다.
*** 정제수율은 사용하고자 하시는 Sample type, species, antibody isotype에 따라 달라질 수 있습니다.

xTractor Buffer를 Capturem™ 컬럼에 첨가하여 Equilibration을 진행 후, His-tagged protein을 첨가하고 결합시킨다. 뒤이어 Washing Buffer를 이용하여 세척 및
원심분리 후 Elution Buffer를 이용하여 용출한다. Miniprep과 96well plate 타입의 경우는 Elution buffer 100ul를, Maxiprep의 경우는 Elution buffer 500ul를
사용하여 90%이상 Elution 가능하다.

장시간을 필요로 하는 레진 정제에 비해, Capturem™ membrane은 표면적과 용량 (Capacity)가 크기 때문에 빠른 시간 내에 막 통과가 가능하므로 단백질 변성이 없는
단백질을 정제 가능하다. 또한 낮은 압력으로 용액이 통과할 수 있으므로 단백질의 품질 저하를 방지하며, Bed volume이 작아 고순도로 정제할 수 있다.

293T cell에 6x His-tagged MAPK1 또는 ADRB2 유전자를 포함한 플라스미드를 Transfection 후, Capturem™ His-Tagged Purification Miniprep Kit을 이용하여 단백질을
정제하였다. Equilibration을 완료한 컬럼에 Lysate 처리, Washing 후 Elution Buffer 300ul로 단백질을 용출하고 4-20% Polyacrylamide gel 전기영동 및 Western blotting을
진행하였다.
Ponceau S staining을 통하여 membrane 내의 모든 단백질이 검출되는 것을 확인할 수 있었으며, MAPK, ADRB2 특이적인 항체를 사용한 Western blot에서 각각의
정제된 단백질을 확인할 수 있었다.
