제품설명

NC (negative control) :
Recombinant baculovirus 감염에 의한 단백질 발현 실험에서 negative control로 사용할 수 있는 baculovirus이다.
일반적으로 baculovirus에 감염되지 않은 세포를 negative control로 이용하지만, baculovirus가 감염된 숙주 세포의 발현 프로파일을 변화시키거나 비감염세포에서는 볼 수 없는 baculovirus 유래 단백질이 존재하기 때문에 비감염세포를 negative control로 사용하는 것을 권장하지 않는다. 또한 Clontech의 BacPAK6 DNA를 이용한 recombinant baculovirus를 감염시킨 세포는 β-galactosidase를 발현하고, wild type baculovirus는 감염세포에서 polyhedrin을 대량 발현하므로 negative control로 적합하지 않다. 따라서 본 NC (negative control) 제품을 baculovirus 감염에 의한 단백질 발현 연구의 negative control 실험에 이용할 수 있다.

GC (Green Control) :
GC (Green Control)는 Aequorea Victoria 유래 green-fluorescent protein (GFP)를 발현하는 baculovirus로, 바이러스 증식의 모니터링이나 배양 조건의 최적화, 역가측정 등에 이용하는 경우의 positive control로 사용할 수 있다.

GCP (Green Control Plasmid) :
GCP (Green Control Plasmid)는 AcMNPV polyhedrin promoter의 조절하에 Aequorea Victoria 유래 green-fluorescent protein (GFP) 유전자를 코딩하고 있는 transfer vector이다. ProEasy 시리즈, GFP를 포함하지 않는 ProFold 시리즈 (ProFold™-0, ProFold-PDI, ProFold-PDI*)Clontech BacPAK6, BD BaculoGold 등의 AcNPV 유래 linearized baculovirus DNA와 함께 곤충세포 내로 co-transfection 하고 polyhedrin 부위의 homologous recombination으로 GFP를 발현하는 recombinant baculovirus를 제작하여 positive control로 이용할 수 있다.

각 baculovirus를 Spodoptera frigiperda 유래 Sf9 세포 감염시킨 후 60시간 후에 회수하여 50 mM Tris-HCl(pH8.0), 150 mM EDTA, 0.5% NP-40으로 lysis하여, 불용성 구획은 30,000×g 15 분 원심분리로 제거하였다. 수용성 단백질을 11% SDS-PAGE에서 분리하여 Coomassie blue으로 염색하였다.
