RiboGone-Mammalian kit는 human, mouse, rat의 total RNA 샘플로부터 5S, 5.8S, 18S, 28S nuclear rRNA (Ribosomal RNA) 뿐만 아니라 12S mtRNA (mitochondrial RNA)를 제거하는 시약이다. 특히, RiboGone은 FFPE 샘플과 같이 degradation된 샘플뿐만 아니라 낮은 quality의 RNA 샘플에도 적용 가능하다. RiboGone-Mammalian Kit는 random-primed cDNA synthesis와 RNA-seq 또는 transcriptom 분석 실험 전, sample preparation 과정에 적합하다 (including the SMARTer Stranded RNA-Seq Kit (634836), the SMARTer Universal Low Input RNA Kit for Sequencing (634938), and the SMARTer Universal Low Input RNA Library Prep Kit (634947)) (그림 1).



그림 1. RiboGone - Mammalian is specifically designed to work with low input RNA samples containing between 10-100 ng of full-length or degraded total RNA. Samples processed using the RiboGone kit are ready for cDNA synthesis with any random-primed SMARTer RNA-Seq kit (including the SMARTer Stranded RNA-Seq Kit, the SMARTer Universal Low Input RNA Kit for Sequencing, and the SMARTer Universal Low Input RNA Library Prep Kit).

그림 3. Sequencing data of RiboGone - Mammalian treated RNA samples highly correlate with MAQC qPCR data for the same reference RNA. The high level of correlation between the RNA-Seq and MAQC data (R=0.860) shows that depletion of rRNA with RiboGone does not interfere with detection of other genes by RNA-seq.
